     Abstract Background and objective: Primary hypertension is the major cause of male reproductive dysfunction. L-NAME induced hypertension is widely used to represent primary hypertension in human since it decreases NO production and increases oxidative stress. Tangeretin is a flavonoid found in citrus fruit peels as well as orange juice. Interestingly, little information regarding the effect of tangeretin on reproductive dysfunction has been scientifically tested in hypertensive animals. Thus, the objective of this study was to investigate the effect of the tangeretin on reproductive dysfunction in male hypertensive rats. Methods: Male Sprague-Dawley rats were induced to be sustained hypertension by Nω-nitro-L-arginine methyl ester hydrochloride (L-NAME 40 mg/kg) in drinking water for 5 weeks. The hypertensive rats were treated with tangeretin at dose 15 or 30 mg/kg or sildenafil (10 mg/kg) for the last two weeks. After each treatment, mean arterial pressure (MAP), intracavernosal pressure (ICP), oxidative stress biomarkers were analyzed. Furthermore, eNOS protein expression in penile tissue was evaluated. Sperm concentration and motility were counted. In addition, histomorphometric evolutions of seminiferous tubules were tested. Results: L-NAME rats showed high blood pressure associated with decreased ICP/MAP ratio, eNOS protein expression. Furthermore, alterations of sperm quality and testicular morphology related to high level of systemic and local MDA. Tangeretin significantly reduced MAP and improved erectile response to cavernous nerve stimulation and restore eNOS expression in L-NAME rats (p<0.05). Tangeretin and sildenafil significantly increased sperm concentration and sperm motility, attenuated histomorphometric changes of seminiferous tubule and oxidative stress in hypertensive rats in comparison with control testes (p<0.05). Conclusions: These findings cloud suggest that tangeretin reduces blood pressure, improves male reproductive dysfunction in NO deficient hypertension. These effects are likely to related with its antioxidant property, along with increasing NO bioavailability. Keywords: Tangeretin, L-NAME, Oxidative stress biomarkers
