     บทคัดย่อ The present study aimed to investigate the efficacy of disinfectant solutions that commonly used in hospitals against Acanthamoebae cysts (genotypes T3, T4 and T5). After incubation with disinfectant, the cyst viability was examined by staining with 0.4% trypan blue.The result showed that treatment with 1%Virkon® was efficient against T4 and T5 genotypes for 10 min. Whereas T3 genotype had to use 2% Virkon® for inactivation. In this study, Clorox® solution with 2.5% and 6% sodium hypochlorite was not efficient. Exposure to 70% ethanol for 30 min was effective for T4 and T5 but not effective for T3, whereas 95% ethanol can kill all genotypes for 30 min. The results showed that T3 genotype is more resistance than the other two genotypes (P< 0.0001) and 2% Virkon® had highly effective (P< 0.005). The viability and non-viability after treatments were determined by staining with 0.4% trypan blue and confirmed by NNA (non-nutrient agar) culture. For heating, all genotypes were not completely inactivated at 55°C, whereas exposure at 65 °C did result in inactivation. Effective disinfection is important not only to prevent the transmission of Acanthamoeba itself but also to reduce the risks of infection with other microbial pathogens that are associated intracellularly with Acanthamoeba.
