บทคัดย่อ
การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาแบบจำลองการสร้างรอยผุสำหรับใช้ประโยชน์ในงานวิจัยทางทันตก
รรม ด้วยเชื้อก่อโรค 2 สายพันธุ์ คือ สเตร็ปโตค็อคคัส มิวแทนส์และแลคโตบาซิลลัส อะซิโดฟิลลัส ให้เจริญเติบโต
ร่วมกันบนชิ้นเนื้อฟันจำนวน 4 ชิ้น เป็นระยะเวลา 1 3 และ 5 วัน โดยมีชิ้นเนื้อฟันปกติเป็นกลุ่มควบคุม เลี้ยงใน
อาหารชนิดเหลว De man Rogosa and Sharpe (MRS) ร่วมกับซูโครสร้อยละ 5 บ่มด้วยอุณหภูมิ 37 องศา
เซลเซียส คาร์บอนไดออกไซด์ร้อยละ 5 จากนั้นสังเกตลักษณะไบโอฟิล์ม วัดความลึกของ รอยผุและวิเคราะห์
ปริมาณแร่ธาตุโดยการทดลองจะถูกทำซ้ำ 3 ครั้ง พบความลึกของรอยผุ 51 143 และ 186 ไมโครเมตร ในวันที่ 1
3 และ 5 ตามลำดับ เริ่มพบไบโอฟิล์มได้ตั้งแต่วันแรกของการแช่ในสารละลายแบคทีเรีย โดยจะมีปริมาณมากขึ้น
และหนาขึ้นเมื่อเวลาผ่านไป อัตราส่วนระหว่างแคลเซียมต่อฟอสฟอรัส คือ 1.69 1.30 และ 0.84 ในวันที่ 1 3 และ
5 ตามลำดับ ซึ่งทุกกลุ่มมีค่าน้อยกว่ากลุ่มควบคุม ดังนั้นผลการศึกษาจากแบบจำลองด้วยเชื้อก่อโรคนี้จึงสามารถ
สร้างรอยผุบนชิ้นเนื้อฟันมนุษย์ได้ และนำไปประยุกต์ใช้ในงานวิจัยทางทันตกรรมต่อไป

Abstract
This study aimed to develop a microbial model to create caries-like lesions to be used
in dental research. Two common cariogenic bacteria (S. mutans and L. acidophilus) were cocultured and grown on four human dentin slices for 1, 3, 5 days and sound dentin slices were
used for control. The bacteria were cultured in a sterile De Man Rogosa and Sharpe (MRS) broth
with 5 % sucrose, incubated at 37˚ c in an atmosphere of 5 % CO₂. After the experimental
periods, lesion depth, biofilm appearance and mineral content were determined. This
experiment was repeated three times. Caries-like lesions depth were found 51, 143 and 186 µm
on day 1, 3 and 5, respectively. Biofilm was found on the first day, it was increased by both
number and thickness over time. Ca/P ratio was 1.69, 1.30 and 0.84 in day 1, 3 and 5,
respectively. Ca/P ratio of all periods were lower when compared with the control group. This
microbial model has developed caries-like lesions on dentin and can be used for future research.
