Alteration of miR-451a responding to vitamin C supplementation in poorly

บทคัดย่อ |
Type 2 diabetes mellitus (T2DM) is a worldwide health problem and rapidly increases in
prevalence. Low levels of anti-oxidant such as vitamin C in T2DM patients have been reported
and these were accompanied by high levels of lipid peroxidation. Supplementation with
vitamin C has been shown to have beneficial effects on oxidative condition and metabolic
system as well as immune status, especially, in poorly controlled T2DM. To gain insight into
the effect of vitamin C at molecular mechanisms, we examined circulating microRNAs
(miRNAs), g group of small RNA function as a regulator of gene expression, in response to a
1,000 mg vitamin C supplementation daily for six weeks in T2DM patients. Five participants
who had increased vitamin C level, improved oxidative status and polymorphonuclear
function were selected to screen for miRNA expression by compared between pre-and postvitamin C consumption using miRNA microarray and confirmed by qRT-PCR. The results
demonstrated that miR-451a was significantly decreased after vitamin C supplementation (-
3.29-fold decrease, P=0.03). Bioinformatics analysis revealed target genes involving signaling
pathways related to those conditions suggested that the miRNA may play a role in T2DM and
may be related to increased anti-oxidative activities and polymorphonuclear function. These
data indicated that consumption of vitamin C altered miRNA expression and the functional
role of this miRNA in uncontrolled T2DM patients awaits further studies. |
