     Abstract Our previous developed melatonin niosome gel using poloxamer407, a thermosensitive polymer with an advantage in facilitating fluid state at 25˚C and gel state at 37˚C, was not able to differentiate adhesiveness by using mechanical testing. ATR-FTIR, a highly sensitive method for detecting chemical interaction, was interested for use to elaborate the effect of temperatures on chemical interaction between mucosa and the melatonin niosome gel. In experiment, excised mucosa of porcine esophagus was applied with blank niosome gel which composed of poloxamer407, polyvinyl pyrrolidone and hydroxypropyl methylcellulose, and melatonin niosome gel, at controlled temperatures of 25±2˚C and 37±2˚C for 1 and 5 min, and then analyzed by ATR-FTIR. The results showed that at 25 ˚C, a peak at 1551 cm-1 (N-H) was identified with samples of the mucosa treated by melatonin niosome gel. At 37˚C, peaks at 1640 and 1551 cm-1 for C=O and N-H, respectively, observed with melatonin niosome gel, while 1551 cm-1 with blank niosome gel. Both treatments with melatonin niosome gel and blank niosome gel for 1 and 5 min gave no difference, on FTIR spectra of the mucosa at 37˚C, representing body temperature. In conclusion, there were changes in C=O and N-H interactions between the mucoadhesive gel and the mucosa rapidly occurred at body temperature, which could lead to explain the adhesive effect of the gel on the mucosa.
