A Comparative Study of 20U and 50U of Intraoral Abobotulinum Toxin A (Dysport®) Injection
in Post Traumatic Trigeminal Neuropathic Pain Patients |

Background: Botulinum Toxin A(BTX-A) has a growing evidence of its effectiveness in several neuropathic condi- tions such as post traumatic trigeminal neuropathic pain (PTTNP). However, studies for optimal dose and injection technique for its analgesic effect is still lacking.
Objectives: To investigate the efficacy and safety of 2 doses of BTX-A in patients suffering with PTTNP.
Methods: Thirteen PTTNP patients were enrolled in a 12-week, double blinded, randomized dose response trial of 2 fixed doses (20U and 50U) of BTX-A(Dysport®). Pain intensity was the primary outcome measured by visual analogue scale. Safety and tolerability were measured based on participant’s self-report using a daily pain diary. Mann-Whitney and Fischer’s Exact test were used for data analysis.
Results: Sixty-nine percent (n=9) of participants had significant pain reduction of ≥30% from baseline pain. Fifty-four percent of the total sample have experienced mild and transient side effects. There was no significant difference between 20U and 50U of BTX-A in the reduction of pain intensity. In addition, there was no association between dosage and the duration of analgesic effect and the occurrence of side effects.
Conclusions: BTX-A is a potential alternative treatment for PTTNP patients. These results suggest that a low dose of BTX-A for intraoral injection is sufficient and safe for pain relief.
