Analysis of Active Ingredients of Thai Propolis Extracts Using High Performance Liquid Chromatography |

บทคัดย่อ |
Background: Propolis has been proven to have numerous beneficial effects in dentistry, such as an effective pulp-capping material, intra-canal medicament, and in preservation of periodontal ligament cells. These are attributed to flavonoids and phenolic compounds.
Objectives: To determine the presence of vicenin-2, vitexin, apigenin, and cinnamic acid in Thai propolis extracts using high performance liquid chromatography (HPLC); and, to analyze whether its concentration will be affected by the ethanolic concentration used in extraction and its source location.
Materials and Methods: Fifty milligrams of propolis from Nong Khai and Loei were extracted using either 100%,90%,80%, or 70% ethanol. These were analyzed using HPLC with a mobile phase gradient system of 10-100% acetonitrile in 0.05% aqueous ortho-phosphoric acid, flow rate of 0.8mL/min, and detection wavelength of 280nm. The concentration of the four components were compared according to location and ethanolic extraction using Two-way ANOVA with significance level at p<0.05.
Results: The source of propolis and the percentage of ethanol used in its extraction both have a significant role in the concentration of all active ingredients (p < 0.01). Vicenin-2 is significantly affected by each change in the percentage of ethanol extraction. Vitexin and cinnamic acid showed statistical difference between 100%,80% and 70% ethanolic extracts, respectively. Apigenin results in significant difference between 100%, 90%, and 70% ethanolic extracts only.
Conclusion: Propolis from Nong Khai have a significantly higher amount of the four active ingredients as compared to Loei samples. These ingredients have significantly higher concentrations in 70% ethanol extraction, as compared to higher concentrations of ethanol.
