     Abstract Hippocampal neurogenesis is continuously generated throughout life, but it is markedly declined with age. D-galactose (D-gal), a monosaccharide, enhances brain aging through oxidative stress accumulation resulting in neuroinflammation, neurodegeneration and memory impairment. Caffeic acid, a non-flavonoid phenolic phytochemical, acts as an antioxidant via interrupting the formation of free radical and decreasing of oxidative stress in the brain. Therefore, the current study was designed to assess the protective effect of caffeic acid on impairment of memory and hippocampal neurogenesis in aging rats caused by D-gal. Fifty-four Male Sprague-Dawley rats were divided into 6 groups. Rats received D-gal (50 mg/kg) by i.p. injection with/without caffeic acid (20 or 40 mg/kg) by oral gavage for 8 weeks. Memory was evaluated using novel object location and novel object recognition tests. Hippocampal neurogenesis was investigated using 5-bromo-2’-deoxyuridine (BrdU)/neuronal nuclear protein (NeuN) immunofluorescence staining to determine cell survival related to mature neurons. The results found that D-gal administration induced memory impairment and depletions in cell survival related to mature neurons. However, co- administration with caffeic acid both 20 and 40 mg/kg attenuated these impairments. These results reveal that caffeic acid could protect against reduction of neurogenesis associated with memory in brain aging.
