The present study aimed to investigate the effect of melatonin on 5-fluorouracil (5-FU) - induced oxidative stress that interferes with hippocampal neurogenesis and antioxidant enzyme and protein expression levels in the rat hippocampus and prefrontal cortex. Rats were administered with 5-FU (25 mg/kg, i.v.) on day 9, 12, 15, 18 and 21 or received melatonin (8 mg/kg, i.p.) from day 1 to day 21 of the experiment. The results postulate that melatonin could protect against the side effects of 5-FU on a reduction of hippocampal neurogenesis by preventing cell cycle arrest and up-regulating levels of antioxidant enzymes and proteins related to regulation of neurogenesis in the hippocampus and prefrontal cortex, which plays an important mechanism on neurogenesis and memory formation. |
