Abstract
Background and Objective: O-GlcNAcylation is an important O-linked glycosylation of nucleocytoplasmic proteins with a single molecule of N-acetylglucosamine. Aberrant O-GlcNAcylation was found to promote the development and progression of cancer. In this study, we have explored the role of O-GlcNAcylation in melanoma progression.
Methods: O-GlcNAcylation in MNT-1, a malignant melanoma cell line, was suppressed by a specific siRNA against N-acetylglucosaminyltransferase (OGT), compared with siControl. Cell proliferation and drug sensitivity were measured by SRB assay. Western blotting and immunofluorescent staining were used to determine the level of O-GlcNAcylated proteins (OGP). The molecular mechanism was explored by Western blotting.
Results: The level of OGP in MNT-1 was drastically reduced after treatment with siOGT compared with control siRNA (siControl). Knockdown of OGT significantly suppressed the cell proliferation ability of MNT-1 via inhibition of the Akt signaling pathway. The drug sensitivity assay revealed the synergistic effect of siOGT with cisplatin against MNT-1 melanoma cells, suggesting the potential of using OGT suppression for cisplatin sensitization for melanoma treatment.
Conclusions: In conclusion, we have provided evidence to improve melanoma therapy via targeting O-GlcNAcylation, which is identified to be involved in melanoma cell proliferation and chemoresistance.
