     Abstract Cholangiocarcinoma (CCA) is a primary malignant tumor arises from biliary tract epithelia either within the liver or the biliary tract. CCA is a major public health problem in the Northeast of Thailand with the highest incidence and mortality rate of the world. Because of lacking early symptoms and range of possible alternative diagnosis, patients with CCA usually present lately with an advanced stage, which is less successful treatment. The CCA intact cell and its products can be exfoliated and pass through bile duct and found in the bile. Consequently, the novel cancer biomarkers can be discovered in the bile and might be detected in the feces if it can pass through the gastrointestinal tract. All Bile 8 cases of 2 Mass forming types, 2 Periductal infiltrating types, 2 Intraductal growth types and 2 non-cancer control were labeled with isobaric tags using Isobaric tags for relative and absolute quantitation (iTRAQ®) to determine the relative abundance of bile proteins identified by liquid chromatography-mass spectrometry (LC-MS/MS). The results were validated with label-free quantitative acquisition, sequential window acquisition of all theoretical fragment-ion spectra (SWATH®). A total of 1240 unique proteins were identified and eighty-five proteins were elevated in CCA compared to control group, including several candidate CCA biomarkers: clusterin, hemopexin, alpha-1-antitrypsin, ICAM-1, myeloblastin, osteopontin, cystatin-c, and vitronectin. Developing detection kit with antibody based techniques has been planned.
