     Abstract The aim of this study was to study of growth and lipid accumulation of Chlorella sp. KKU-S2 under mixotrophic cultivation in Bristol’s medium by using molasses as carbon substrate. Effect of different concentrations of molasses and sodium nitrate was performed by batch cultivation. The result presented that the highest growth was found with biomass concentration of 9.23 g/L when cultivated on medium supplemented with 10g/L molasses and 5g/L sodium nitrate, while lipid content of 12.05 % of dried cell weight was found on medium supplemented with 20g/L molasses and 5g/L sodium nitrate. Then, fed-batch cultivation was investigated with initial molasses and sodium nitrate of 20g/L and 5g/L, respectively, then molasses and sodium nitrate was intermitted fed every 2 days of cultivation time, a biomass concentration of 10.88 g/L with lipid yield of 0.704 g/L was obtained at day ten of cultivation with continuous illumination for 24 hours.
