Abstract Bio-hydrogen production from cassava starch wastewater by buffalo dung was conducted in continuous stirred tank reactor (CSTR) under optimum conditions (NaHCO3 of 2.90 g/L, initial pH of 6.76 and COD/TKN ratio of 42.27) obtained from batch experiment. The hydraulic retention time (HRT) was varied from 72, 60 to 48 h. After the operation at HRT of 72 h for 14 days using the optimum pH of 6.76, the methane was detected in system. This was due to the fact that a bio-hydrogen production in CSTR was longer than in batch fermentation. Hence, the methanogenic bacteria became dominant resulted in methane production in the system. In order to overcome this problem, the initial pH was adjusted to 5.5. After pH adjustment, it was found that the optimum HRT was 60 h in which the highest hydrogen content, HPR and HY of 33%, 834 mLH2/L.d and 16.6 mLH2/g-CODadded, respectively, were achieved. The main soluble metabolite product was butyric acid of 72.84% indicating the butyrate type fermentation.
