Abstract Plaunoi (Croton stellatopilosus Ohba.), a Thai medicine plant, has been used for the treatment of various diseases, especially peptic ulcer and related gastrointestinal diseases. The aim of this study was to investigate anti-proliferative and apoptosis induction activities of Plaunoi leaf powder water extract in colon cancer cell lines (HT29 and HCT116 cells) and a non-cancer cell line (Vero cells) by MTT and Vybrant Apoptosis assays, respectively. The water extract inhibited proliferation of HT29 and HCT116 cells at exposure times of 24, 48, and 72 h with IC50 values of 403.83 ± 3.40, 371.26 ± 2.17, 326.03 ± 3.04 µg/ml and 461.34 ± 4.17, 379.66 ± 20.99, 274.97 ± 21.57 µg/ml, respectively. In contrast, the water extract inhibited the growth of the non cancer Vero cells with IC50 values of 841.14 ± 19.54, 765.93 ± 37.34, 733.24 ± 5.32 µg/ml at exposure times of 24, 48, and 72 h, respectively. The water extract at concentrations of 100, 200 and 400 µg/ml induced apoptosis of colon cancer cell lines (HT29 and HCT116) in a concentration dependent manner but induced less apoptotic cell death in Vero cells.
