     Abstract Flu infection is still the major public health problem worldwide caused by influenza A virus. Animal base compound has used as traditional remedy for many diseases. The highlight of crocodile as source of this study because it has been one of animal evaded extinction, by still maintain anciant shape. Crocodile lymphocyte show anti-bacterial broad-spectrum. Crocodile serum can inhibit virus such as, west-nile virus and HIV. Moreover, there is an evidence that crocodile complement showed a strong innate immune defense. This is the study of immunomodulatory and antiviral functions of Crocodylus siamensis’ liver extract (CLE) in innate immunity genes on A549 cell. CC50 was assayed by MTT to evaluate the harmful to A549 cell. The mRNA expression of cytokine gene was measured by quantitative RT-PCR. We found that, CLE 32.01 mg/ml toxic to 50% cell viability. At 39.11 ug/mL inhibit 50% of AH3N2 replication. The CLE 0.35 mg/ml at the incubation time 24 h on A549 cell can stimulate IL-2, Mx-1, CCR5/RANTES and CXCL10/IP10 up regulated in A549 cell with CLE treatment. IL-2 is up regulated in AH3N2 infection with CLE treatment. We can conclude that CLE show potential to promote immune response of the cell. The possible mechanism will be evaluated further.
