     Abstract Staphylococcus aureus membrane fatty acids are generally contained a mixture of branched-chain fatty acids (BCFAs) and straight-chain fatty acids (SCFAs) which the BCFA function is an increasing membrane fluidity and SCFAs decrease membrane fluidity. The objective of this study was to determine the cellular fatty acid methyl ester profile among clinical S. aureus isolates including vancomycin-susceptible S. aureus (VSSA), heterogeneous vancomycin-intermediate S. aureus (hVISA) and vancomycin-intermediate S. aureus (VISA). Thirty S. aureus isolates were cultivated on media with and without vancomycin before conducted to extract and analyzed the cellular fatty acids by Gas chromatography with flame ionization technique. This study showed that vancomycin had a significant impact on cellular fatty acids level of S. aureus. The membrane fluidity of VSSA cells had reduced a total level of SCFAs (p<0.05) and increased a total level of BCFAs under vancomycin stress (p<0.05). However, vancomycin can induce the change of only BCFAS but no effect on SCFAs in hVISA while vancomycin had no effect on both BCFAs and SCFAs in VISA cells. Therefore, we concluded that vancomycin has much impact on VSSA followed by hVISA, but no effect on VISA cell.
