     Abstract A plethora of evidences have shown that HDL can be used as a biomarker in the diagnosis of several diseases. Various publications showed that isolating HDL requires a large sample volume. Thus, the limited sample volume sometimes becomes a problem in research and diagnostic needs. This study aimed to isolate HDL from serum samples with small volume variations of 200 and 500 µL. HDL was isolated using the HDL Purification Kit (Cell Biolabs) and continued with the measurement of protein concentration using the BCA protein assay. To ensure the success of HDL isolation, ApoA-1 was used as an HDL biomarker and detected using Western Blot. The results showed HDL isolated from 200 and 500 µL of serum had protein concentration of 7.125 and 9.375 µg / µL respectively and they showed the presence of ApoA-1 (~29 kDa). It can be concluded that this method can be used as an alternative method to isolate HDL with a small sample volume.
