| บทคัดย่อ |
Orchids are highly important and valuable plants due to their diversity and the beauty of their flowers. Nowadays, there are many orchid species going to extinct because of environmental changes and over-collection for commercial uses. Cryopreservation is an alternative method for storage of plant genetic resources. The aim of this study was to evaluate the effectiveness of vitrification method for cryopreservation of Rhynchostylis gigantea seeds. The seeds were pre-cultured for 24 h in a liquid ND medium supplemented with 0.5M sucrose, incubated with a mixture of 0.4M sucrose and 2M glycerol for 30 min and finally dehydrated with PVS2 for 0-120 min at 25°C prior to storage in LN for 24 h. The orchid seeds successfully preserved by vitrification method. The results found that the optimal dehydrated time in PVS2 was 30 min, giving 61% of the highest recovery rate after culturing the seeds on ND medium for 30 days. The results of genetic stability assessment revealed that RAPD patterns were similar between cryopreserved and non-cryopreserved seeds. A total of 92.5% of RAPD primers gave the highly SI value which was ranged between 0.9 and 1.0 indicating that the genetic stability of seeds stored in liquid nitrogen was maintained. |
