     Abstract Tyrosinase inhibitor plays a key role for skin whitening cosmetic and hyperpigmentation disorders treatment. Although, several well-known inhibitors are available, their cytotoxicity and mutagenicity were observed. Therefore, the new compounds with the effectiveness and nontoxicity are needed. In light of this background, the current research was aimed to evaluate tyrosinase inhibitor tetrapeptide modified from C-terminal cysteine heptapeptides, Leucrocin I derivative. The result found that among all of 8 tetrapeptides, T7 exhibited higher tyrosinase inhibitory activity than its template and showed the highest diphenolase inhibitory activity. Therefore, it was selected for further investigation in its inhibitory mechanism and enzyme inhibitory kinetic study. Kinetic study exhibited that the T7 is competitive irreversible inhibitor. Molecular docking indicated that the T7 directly bound to active site of tyrosinase using hydrogen bond and electrostatic interaction. In addition, proximate binding of -SH group of cysteine residue in the T7 oriented to the copper atom possibly suggested the coordinate covalent bond which caused the irreversible inactivation. The copper chelation activity of T7 was found and confirmed this covalent formation. This result suggested that the T7 was highly possible to be developed as tyrosinase inhibitor for cosmetic and hyperpigmentation treatment.
