     Abstract Background: Cholangiocarcinoma (CCA) is the second most common primary liver cancer and, CEA and CA19-9 often use for CCA routine diagnosis, but they are low sensitivity and low specificity. Valosin-containing protein (VCP), is a critical mediator of protein homeostasis, overexpression of VCP occurs in many carcinomas, and often in correlating with the progression, prognosis, and metastatic potential of carcinoma, the specific function of VCP in human CCA remains unclear. In our previous study, VCP is a candidate mitochondrial protein which expressed only in papillary CCA tumor. This research aims to study VCP as a diagnostic marker in CCA. Material and Methods: To study the expression of VCP in CCA papillary tissue and nonpapillary tissue was analyzed by using the intensity of mass spectrometry and western blot, respectively. Using immunohistochemistry (IHC) at Human Protein Atlas was compared to the expression of the VCP between CCA and adjacent normal tissues. To predict VCP as a secreted protein via the classical pathway or non-classical pathway, SecretomeP software by using NN-score >0.5, SignalP software by using likelihood ratio, and European Molecular Biology Open Software Suite (EMBOSS) by using the value of the minimum weight at 3.5 were used to predict signal peptide cleavage sites of VCP sequence. Finally, using the Plasma Proteome Database (PPD) to predict VCP is presented in the plasma or serum. Results: Based on the Human Protein Atlas, results of IHC revealed that the expression of VCP in CCA tissues was higher than that of those in adjacent normal tissues. VCP was also the highest expression in all of four mitochondrial proteins in only the papillary CCA tissue by using signal intensity from the mass spectrometry. However, western blot results indicated that VCP expressed in both papillary and nonpapillary CCA tissues. EMBOSS software predicted that there were two signal peptide cleavage sites in the VCP sequence. Also, we found that VCP can be present in the plasma or serum as well by using PPD. Conclusion: The significant expression of VCP in CCA tissue was demonstrated, and we can be found VCP in plasma or serum by using bioinformatics tools, which might be a candidate diagnostic biomarker of CCA patients. Therefore, VCP should be further investigated in the serum of CCA patients.
