Research Title Determination of major bioactive compounds from Derris scandens using HPLC-UV

     Abstract Derris scandens is woody climbing vine plant in Thai called “Thaowan Priang” (Hog Creeper Vine). This plant is used for analgesic and anti-inflammatory activities. D. scandens is widely used to treat muscle pain in form of dry extracted capsule. D. scandens has various constituent compounds, most of them are isoflavonoids and their derivatives. Thai Herbal Pharmacopoeia (THP) described genistein-7-O-[α-rhamnopyranosyl-(1 to 6)-β-glucopyranoside] (GTG) and genistein for chemical markers. The other isoflavonoids in this plant included lupalbigenin and 6,8-diprenyl genistein can be used as chemical markers for quality control of D. scandens. In this experiment, we aim at development of high-performance liquid chromatography using UV detectors (HPLC-UV) method for simultaneous determination of major compounds in D. scandens. Our HPLC-UV method used acetonitrile and 0.05% ortho phosphoric acid for gradient mobile phase with reverse phase C-18 column and assay time is one hour. Our method can detect four major compounds at 280 nm and quantitative analysis range of GTG, lupalbigenin, and 6,8-diprenyl genistein were 0.312 - 40.0 µg/ml and range of genistein was 0.312 – 5.0 µg/ml. Intra- and inter-day precision was 0.13–3.02% and 0.42–6.79%, respectively. The accuracy represented by recovery of authentic GTG, genistein, lupalbigenin and 6,8-diprenyl genistein spiked into samples were 93.33-109.63%. The limit of qualification of GTG, genistein, lupalbigenin and 6,8-diprenyl genistein were 182.19, 25.15, 45.07, 163.14 ng/ml, respectively. The limit of detection of GTG, genistein, lupalbigenin and 6,8-diprenyl genistein were 54.66, 7.54, 13.52, 48.94 ng/ml, respectively. This method validation can confirm amount of four compounds that found in D. scandens. The developed method was applied to determine level of these four compounds in stem of D. scandens. The amount of GTG, genistein, lupalbigenin and 6,8-diprenyl genistein in stem extracted contained 16.63 ± 0.58, 1.69 ± 0.07, 90.32 ± 4.40 and 100.51 ± 4.83 µg/mg, respectively. This method is useful for detection of bioactive compounds in plant and derived products.
