     Abstract Lymphatic filariasis, caused by Brugia malayi is endemic in Thai-Malaysia border and cats serve as the important reservoir host. On the other hand, in natural survey domestic cats hold B. pahangi infection. Microfilarial differentiation of B. malayi and B. pahangi by morphology using microscopic examination is difficult. The method is laborious, tedious and time consuming, and carry a low sensitivity and a need for specially trained personnel. Here, we developed real-time fluorescence resonance energy transfer (FRET) PCR combined with melting curve analysis for differentiated detection between B. malayi and B. pahangi DNAs in host blood using one set of primer and hybridization probes specific for HhaI repetitive DNA for B. malayi (GenBank accession no. M12691). The real-time FRET PCR can be differentiated between B. malayi microfilaria inoculated human blood and B. pahangi microfilaria in infected cat blood by melting temperature (Tm). The mean Tm values SD of the B. malayi and B. pahangi were 56.12 ± 0.01 and 52.75 ± 0.03 respectively. Melting curve analysis produces rapid, accurate, and sensitive alternative for differential detection of both species of brugian filaria in host blood, allows high throughput and can be performed on very small samples. The method therefore has great potential for application in epidemiological studies in human, animal reservoir hosts and mosquito vectors.
