Introduction: The oxidant/antioxidant balance is suggested to be one of a significant factor for progression and recurrences of ulcerative colitis (UC). The present study aimed to investigate dextran sulfate sodium (DSS)-induced UC in mice via oxidative stress pathway. Materials and methods: UC was induced in adult male ICR mice by daily oral administration of DSS (40 kDa) solution (6 g/kg/day) using gastric lavage for 4 and 7 days (n=4). The control mice were simply given drinking water (1 mL/day). After the end of induction, colon was collected to determine expressions of superoxide dismutase 1 (Sod1), superoxide dismutase 2 (Sod2), and catalase (Cat) mRNA by RT-qPCR. Results: DSS significantly suppressed the expression levels of Sod1 and Sod2 mRNA compared to the control at both day 4 and day 7 of the induction. The expression of Cat mRNA was significantly down-regulated in DSS-treated mice on the day 7 compared to the control. Conclusion: These findings assured the pro-oxidant potential of DSS to induce UC in ICR mice by decreased mRNA expression of Sod1, Sod2, and Cat. It is of interest for further study that DSS-induced colitis in ICR mice on oxidative/antioxidant balance.
